CRLX101 Selected for Oral Presentation at 17th Annual Symposium on Anti-Angiogenesis and Immune Therapies for Cancer

Cerulean Pharma Inc. (Nasdaq: CERU), a leader in Dynamic Tumor Targeting, announced today that preclinical data for CRLX101 as monotherapy and in combination with Avastin(R) have been selected to be the subject of an oral presentation at the 17th Annual Symposium on Anti-Angiogenesis and Immune Therapies for Cancer to be held February 19-21, 2015 in San Diego, California.

Elizabeth Pham, Ph.D., Postdoctoral Fellow, Sunnybrook Research Institute in Toronto, will discuss the results of preclinical mouse models of advanced metastatic cancer in an abstract presentation titled Translational evaluation of the investigational nanoparticle-drug conjugate CRLX101 with or without bevacizumab in advanced ovarian and breast cancer, as part of a session on biomarkers on Friday, February 20, 1:25 p.m. PT. The ovarian data were the subject of a manuscript published in Clinical Cancer Research.

In an ovarian cancer model, CRLX101 monotherapy was highly efficacious, and chronic low-dose CRLX101 with Avastin reduced HIF-1 up-regulation and resulted in synergistic efficacy, with minimal toxicity in mice. In breast cancer models in mice, CRLX101 monotherapy was highly efficacious, and CRLX101 with Avastin resulted in dramatic, durable primary tumor shrinkages and greatly improved survival.

These data support a growing view in the anti-angiogenesis community that adding a durable HIF-inhibitor like CRLX101 to VEGF inhibitors, which drive up HIF, could lead to important benefits for patients, said Elizabeth Pham, Ph.D.

Elizabeth Pham is a Postdoctoral Fellow in the lab of Robert S. Kerbel, Ph.D., Sunnybrook Research Institute, who serves as a Chair and on the selection committee for the Symposium on Anti-Angiogenesis and Immune Therapies for Cancer. Dr. Kerbel recused himself from the scoring for this abstract due to his affiliation with the research.

About CRLX101

CRLX101 is a dynamically tumor-targeted nanoparticle-drug conjugate (NDC) designed to concentrate in tumors and slowly release its anti-cancer payload, camptothecin, inside tumor cells. CRLX101 inhibits topoisomerase 1 (topo 1), which is involved in cellular replication, and hypoxia-inducible factor-1 (HIF-1), which research suggests is a master regulator of cancer cell survival mechanisms thought to promote drug and radiation resistance. CRLX101 has shown activity in four different tumor types, both as monotherapy and in combination with other cancer treatments. CRLX301 is a dynamically tumor-targeted NDC designed to concentrate in tumors and slowly release its anti-cancer payload, docetaxel, inside tumor cells. In preclinical studies, CRLX301 delivers up to 10 times more docetaxel into tumors, compared to an equivalent milligram dose of commercially available docetaxel and was superior to docetaxel in seven of seven animal models, with a statistically significant survival benefit seen in five of those seven models. In addition, preclinical data show that CRLX301 had lower toxicity than has been reported with docetaxel in similar preclinical studies. CRLX301 is currently in Phase 1 clinical development.
